A Study Comparing The Effects of Single Doses of Sertraline 100mg, Amitriptylint 50mg, and Placebo on Daytime Alertness/Sleepines and Performance in Healthy Young Adults

Abstract
The relatively new antidepressant sertraline, a highly selective inhibitor of serotonin-uptake, is known to be free of sedative side-effect commonly found in conventional antidepressants. This study was conducted to assess the effect of
sertraline
on
some aspects of daytime alertness/sleepiness and performance.
This double-blind crossover study was conducted in 9 healthy young male volunteers adminiatered single doses of sertraline 100mg. amitriptyline 50mg. placebo with each treatment separated by a 7-day washout period. All subjects completed the
multiple
sleep latency test (MSLT) and a battery of cognifive and psycnomotor performance tests (addition, digit span, digit symbol substitution. free recall. choice reaction time, pegboard and finger tapping) at 2, 4, 6, 8, & 10 hours post-dose.
Compared to placebo sertraline significantly increased sleep latency at 6 and 8 hours and the mean over all time points. Sertraline showed no significant change in any performance test relative to placebo. In contrast amithptyline significantly
decreased both the mean sleep latency and at all time points except 2 hours relative to placebo and sertraline. Also. amitriptyline produced significant impairments of performance in addition. DSST and finger tapping.
Tnese fesults indicated that sertraline has no sedative activity. evident from the absence of apparent impairments of cognitive and psychomotor function, but rather posesses alerting properties. This suggests that sertraline may be a safer and
more
suitable antidepressant than amitriptyine and other tricyclics particularly for out-patients who are ambulatory and lead active lives.
